The biology
Arousal sends a signal.
When you are aroused, your body releases nitric oxide and a messenger called cGMP. That signal relaxes blood vessels in the penis, blood flows in, and you get an erection.

An enzyme interrupts it.
An enzyme called PDE5 breaks the cGMP signal down, sometimes too quickly. Blood vessels stay tight, blood flow stays restricted, and the erection does not happen or does not last.
